Methods for microbial screening have advanced greatly in the last few years. In this study, we will investigate a parallel format, plate-based, microbioreactor to perform DO triggered fed-batch experiments to screen E. coli and P. pastoris strains to elucidate which clones are the best 1.) growers and 2.) best growers/producers. We de-risked our larger scale/larger volume, more expensive systems, and obtained the high value experimental parameters at 1mL required for our bio-process scale up. We were able to observe experimental nuances not visible or detectable in our other platforms by cost effectively increasing our data density which provided a better understanding of our fermentation parameters for each individual clone, media, vector. In our experience, the BioLector platform provided the same performance as industrial scale bioreactors.
